Abstract

На основі даних одно- і двомірної 1H-ЯМР-спектроскопії (500 МГц) розглянуто комплексоутворення акридинового забарвника профлавіну із самокомплементарними дезокситетрарибонуклеозидтрифосфатами Sr -Ci(GpCpGpC), 5'-d (CpGpCpG), S'-d (ApCpGpT), 5'-d (ApGpCpT) у водному розчині. Двомірна гомоядерна 1Н-ЯМР-спектроскопія (2M-COSY і 2M-NOESY) використана для повного віднесення сигналів протонів молекул у розчинах та для якісного аналізу характера взаємодії профлавіну з тетрануклеотидами. Розглянуто експериментальні концентраційні залежності протонних хімічних зсувів молекул та різні схеми комплексоутворення забарвника з тетрануклеотидами, що враховують рівновагу асоціатів у розчині. Вивчено відносний зміст комплексів у розчині і виявлено особливості динамічної рівноваги в залежності від послідовності основ в нуклеотидному ланцюгу. Виходячи з одержаних даних зроблена висновок про переважну інтеркаляцію профлавіну в CG-сайти тетрануклеотидів у дуплексній формі. Порівняльний аналіз найбільш вірогідних структур 1 : 2 комплексів забарвника з тетрануклеотидами виконано на основі розрахункових значень індукованого хімічного зсуву та 2M-NOE-спектру
