Інсулін-залежний цукровий діабет: можливості генної терапії

Abstract
В огляді викладено генно-інженерні підходи, які розробляються для лікування інсулін-залежного цукрового діабету (13ЦД).
Висвітлюються дослідження, спрямовані на забезпечення синтезу інсуліну de novo в неспецифічних клітинах за рахунок переносу в них гена інсуліну для заміщення β-клітин підшлункової залози. Проаналізовано поиіук клітин-мішеней та шляхів набуття ними необхідних якостей: чутливості до глюкози і здатності до продукції функціонально зрілого білка in vivo. Розглянуто також можливість лікування ІЗЦД за допомогою переносу генів деяких цитокінів Т-лімфоцитам-хельперам (Тх-клітинам), шр спричинює змінення су б популяційного співвідношення Т-клітинного складу. Таким чином, можна перервати або збалансувати специфічний імунний процес в острівцевих клітинах, який лежить в основі патогенезу діабету І
типу.
