In vitro model system for investigation of DNA double stranded breaks reparation based on using of higher plants protein extracts

Abstract
It has been established the in vitro model system for investigation of DNA double stranded breaks (dsb ) reparation based on using of higher plants protein extracts. The system is based on searching of possibility of homologous recombination between two linear pUC19 DNA fragments. As a result of recombination processes gene lacZ is restored in their structure and recombinants with blue phenotype appear on the media with IPTG and Z-gal. Dsb in plasmid DNA has been provoked using EcoRI and Bgll restriction enzymes. The possibility versions of recombination in proposed system by means of substrate ligation and nonhomologous recombination is disscused.
