New packaging cells based on the: adapted variant of the Rous sarcoma virus

Abstract

We have constructed a new transformation-defective mutant of duck-adapted variant of Rouse sarcoma virus Prague strains subgroup C (td daPr-RSV-C) – based packaging cell line. F18/pJD which produces replication-defective vector with a liter 106 CfU/ml. Deletion of cis-acting sequences of packaging region and E (direct repeat flanking src) in the adapted variant of retrovirus allowed us to obtaine a pool of retrovirus vector without helper virus sequences.
