Abstract

Експресія інгібітори активатора плазміногена-1 (ІАП-1) стимулюється гіпоксією (8 % О2). За цю стимуляцію відповідає ділянка в промоторі IAП-1 (-175/-159), яка містить елемент, що сприймає гіпоксію (ЕСГ-2). З останнім взаємодіє фактор 1, що його стимулює гіпоксія (ФСГ-1). З ділянкою ЕСГ-2 межує ділянка ЕСГ-1, з якою зв'язується раніше не ідентифікований фактор. У даній роботі зроблено спробу ідентифікувати цей фактор і з'ясувати його роль у регуляції експресії ІАП-1. На підставі зсуву специфічної електрофоретичної смуги доведено, що з сайтами ЕСГ, переважно з ЕСГ-1, зв'язується вищерозташований стимулюючий фактор-2а (ВСФ-2а). В первинній культурі гепатоцитів щура надекспресія ВСФ-2а в однаковій мірі пригнічувала експре­сію ІАП-1-специфічної мРНК та білка ІАП-1 за умов як гіпоксії, так і нормоксії. Первинні гепатоцити щура трансфікували двома рекомбінантними ДНК – конструкцією з геном люциферази під контролем ділянки промотору ІАП-1 довжиною 766 або 276 п. о. і векторами, які експресують ВСФ дикого (ВСФ-2а) або мутантного (ΔВСФ-2а) типу. Останній не виявляє ДНК-зв'язуючої та димеризуючої активностей. Котрансфекція з вектором, який експресує ВСФ-2а, пригнічувала активність люциферази у 8 разів порівняно з контролем, де білок ВСФ-2а був відсутнім. Котрансфекція з вектором, який експресує ΔВСФ-2а, не впливала на люциферазну активність. Мутації обох ЕСГ (і в більшій мірі мутація ЕСГ-1) перешкоджали пригніченню контрольованої промотором ІАП-1 люциферазної активності. Інгібіторної дії ВСФ-2а, опосеред­кованої ЕСГ-2, на відміну від ЕСГ-1, не спостерігалося при надекспресії ФСГ-la. Ці результати свідчать про те, що баланс між двома транскрипційними факторами, ВСФ-2а та ФСГ-1 а, що можуть зв'язуватися з суміжними ЕСГ, відіграє певну роль у регуляції експресії ІАП-1 за умов гіпо- та нормоксії.
