Abstract

Сайт-специфічна рекомбіназа Flp, яка кодується 2 мкм плазмідою з Saccharomyces cerevisiae, бере участь в ампліфікації числа копій згаданої плазміди, інвертуючи одну її частину відносно другої під час реплікації плазміди. В роботі розглянуто складний процес зв'язування білка Flp з ділянкою Flp-рекомбінації, який являє собою ряд послідовних стадій впізнавання. Аналіз первинної послідовності фрагмента ДНК ділянки рекомбінації білка Flp, здійснений для з'ясування здатності цієї ділянки до згинання, яке виникає внаслідок зв'язуванням з нею білка Flp, показав невипадковість розподілу в ділянці динуклеотидів, що мають тенденцію до згинання ДНК у бік малого жолобка ДНК. Цілком імовірно, що фрагмент молекули ДНК у ділянці рекомбінації є не «пасивним» субстратом для реакції рекомбінації, а субстратом, що сприяє оптимальному проходженню рекомбінаційного процесу. Очевидно, саме послідовний процес взаємоіндукованої відповідності Flp-рекомбінази та ділянки рекомбінації забезпечує їх хрективне і специфічне зв'язування.
