Роль поліаніонів як конформерів і конкурентів ДНК в організації хроматину

Abstract

Поліаніонні компоненти клітинного ядра, що беруть участь в організації хроматину, можна розділити на два класи. Перший клас перманентних і транзитних конформерів забезпечує відповідно на тривалий і короткий час специфічні конформації фрагментів ДНК та/або особливі стану відповідних ділянок фібрил хроматину. Другий клас поліаніонів виконує роль конкурентів з ДНК за компоненти хроматину. Так звані «фактори збірки хроматину», що зустрічаються в деяких клітинах у вигляді комплексів з молекулами позахромосомного пулу гістонів, є представниками другого класу ядерних поліаніонів. Функціональний сенс утворення зазначених комплексів може полягати в запобіганні конкуренції гістонів за ДНК, яка викликає in vitro збирання або реорганізацію хроматину в нуклеопротеїд, повністю позбавлений гістона Н1 вже при 1,5–2-кратному ваговому надлишку сумарного гістона над ДНК.
