Seryl-tRNA synthetase – a novel autoantigen during systemic autoimmune diseases

Abstract

Sera of patients bearing autoimmune disease (reumatoid arthritis and systemic lupus erithematosus) and sera of clinically healthy donors were examined for the presence of autoantiboaies directed against cytoplasmic autoantigen– seryl-tRNA synthetase. Immunoreactivity of autoimmune sera was investigated by ELISA and dot-analysis methods. It was shown, that autoantibodies against Ser-RS possess lower affinity in comparison with monospecific antibodies against the same antigen. We have examined the influence of autoantibodies on the functional activity of Ser-RS in the aminoacylation reaction. A conclusion have been made that autoantibodies are more active in the inhibition of the enzymatic activity of target antigen then polyclonal antibodies obtained from immunization of laboratory animals.
