Transcription of the synthetic DNA duplexes with repeats of promoter fragments

Abstract
Peculiarities of the in vitro and in vivo transcription of synthetic DNA duplexes containing the repeats of the «ideal» Pribnow box (period 10 bp) and –10th region of trp-promoters (period 16 bp) are studied. It is shown that these polymers can be effectively transcribed in vitro by E. coli RNA polymerase in the presence of heparin. The predominant in vitro transcription initiation sites in these polymers were revealed. It is shown that relatively short synthetic oligonucleotides containing the Pribnow box as bacterial plasmid constituents can initiate the in vivo RNA synthesis like natural promoters.
