The aclion of vitamin E on RNA- and DNA-polymerase activity of rat liver mitochondria

Abstract

The action of tocopherol on RNA- and DNA-polymerase activity of isolated rat liver mitochondria in condition of its joined addition with Ca2+ -ionophor A23187, Ca2+-channel blacker verapamil or protein kinase C inhibitor pliorbol I2-myristate-13-acetate (PMA) were studied. It was found, that tocopherol, A23187 and verapamil effected RNA and DNA synthesis in mitochondria only if added together with fraction of tocopherol-binding proteins. In case of addition of this fraction together with tocopherol and A23J87 or verapamil, the RNA-polymerase activity of mitochondria of normal rats increased compared to assayes in which only tocopherol and tocoplierot-binding proteins were added. Unlike this, joint addition of tocopherol binding protein, A23187 and verapamil caused inhibition of DNA synthesis in mitochondria of normal but not vitamin E-deficient ruts. The addition of tocopherol decreased the action of these compounds. There were found the differences between RNA-polymerase activity of normal and vitamin E-deficient rats in presence of PMA. It was suggested that Ca ions and protein kinase C may be involved in action of vitamin E on RNA synthesis in rat liver mitochondria.
