Капілярна еластовіскозиметрія нуклеотидів еукаріотів

Abstract

За допомогою запропонованого методу капілярної еластовіскозиметрії вивчали структурні особливості надмолекулярних комплексів ДНК (НК ДНК) на різних рівнях їхньої організації у клітинах еукаріотів. Досліджено залежність величин еластов’язкості НК ДНК від часу лізису і іонної сили. Показано, що константи швидкості релаксації нуклеоїдів для різних клітин еукаріотів істотно відрізняються. Еластовіскозиметричні криві титрування цих клітин бромистим етидієм мали біфазний характер аналогічно кривим при седиментації. Показано, що зміна еластовіскозиметричних характеристик нуклеоїдів L1210 зумовлена дією ендогенних протеаз, так як електрофоретичний склад білків нуклеоїдів при цьому залишався постійним. Нуклеази за цих умов експерименту також були не активними. Метод капілярної еластовіскозиметрії виявився досить інформативним і чутливим у вивченні властивостей НК ДНК.
