Abstract

Triton X-100 treatment of nuclei isolated from rat hepatocytes is shown to remove outer nuclear membrane and leads to the activation 20–30-fold of endogenous DNA synthesis. Results of a number of independent approaches in the analysis of products of the synthesis prove that induced synthesis is not the consequence of the initiation of DNA replication. Perhaps, we observe the acceleration of endogenous DNA elongation catalyzed by DNA polymerase a. Nuclei treated with triton X-100 can be used as effective natural template system (permeable for biopolymers) for the measurement of mammalian DNA synthesis catalysed by DNA polymerase α.
