Uptake the mollicute cells of the analogs of antisense oligodeoxynucleolides

Abstract

Processes of absorption and uptake by cells of Acholeplasma laidlawii PG-8, Mycoplasma fermentans PG-18 and Mycoplasma pneumoniae FH of phosphorothioate, phosphorodithioate and methylphosphonate analogs of oligodeoxynucleotides which are co mplementary to «signature» 16S rRNA sequences have been investigated. Distinctions in efficiency of absorption by given cells of phosphorothioate and methylphosphonate analogs of oligonucleotides are found out. So, the level sorption of thioates by the cells of A. laidlawii PG-8 exceeded a share binding with these cells of methylphosponates more than in 5 times. A fact of significant accumulation by the mollicute cells of thio- and dithioate analogs of oligonucleotides is established. The intracellutar concentration of these compounds at all investigated microorganisms exceeded them extracellular in 104 –105 time. On the basis of received of experimental data the assumption of existence in mollicute cells two of various gears of uptake of analogs of oligonucleotides bearing by negative charge in sugar-phosphate backbone and neutral oligonuc-leotide analogs is stated.
