The quantitative estimation of induced DNA interstrand crosslinks in nanomoles interval

Abstract

The method of estimation is based on the comparison of renaturatton degree for DNA samples with a different quantity of the interstrand crosslinks (1C). Atrenaturation of modified animal DNA the complete recovery of duplex structure after the metting occurs if the Quantity of 1C is > 1 per fragment. The decrease of this Quantity to a value <1 per fragment causes the proportional reduction of renaturation degree. The quantity of cross linked nucleotide pairs in DNA complex with an anticancer drug cisplatin may be detected in nanomoles by spectrophotometric method.
