Alpha-amylase gene as a model for secretion vector construction

Abstract

Bacillus amyloliquefaciens A50 alpha-amylase gene was cloned in E. coli and B. subtills cells. The leader part of the gene which includes the signal peptide coding sequence was used for secretion vector construction. One of the vectors obtained was applied for human leukocyte interferon expression in E. coli and B. subtllis. The secretion of interferon and alpha-amylase was shown to differ in these microorganisms. A large amount of interferon in E. coli is found in cytoplasm while in B. subtilis it is found in the membrane fraction.
