THE DNA PROBE CONSTRUCTION WITH USE THE SILVER SALT. 2. THE COLOURING OF THE MONOMER AND THE SINGLE-STRANDED DNA
Summary
Some aspects of adenine modification as a one of DNA base during the preparing of coloured metal-complex is under investigation. After the N-metoxyadenine formation as a result of formaldehyde reaction with the amino/imino groups of the base the lengthing of lateral chain is occured at the expense of ethanolamine and some metoxy groups. The colour of modified adenine is developed after a silvre addition and the ultimate branched metal-complex possess in addition the fluorescent properties. The kind of clour is determined by pH of incubation media, the violet probe with a 10—15 % modification level is obtained in an alkaline solution in the course of 15—20 minutes.
