Revelation and Characterization of Mitochondrial Plasmid-Like DNA of Cotton
Summary
Mitochondrial DNA was extracted from ethylated seedlings of cotton using the methods of differential and equilibrial ultra centrifugation in CsCl-Hoechst 33258 density gradient. By electron microscopy and agarose gel electrophoresis, it was shown that the mitochondrii of cotton belonging to G. hirsutum L. species (variety 108-F) has an addition to basic DNA of high molecular weight, two minicircular DNA of the size of 6.5 and 2.4 Kb. Analysis of the results of .analytical ultra centrifugation and electrophoregrams of the distribution of the products of segregation of DNA of the cell organelles using restrictase EcoRI showed that the plasmid-like DNA has mitochondrial origin and that it does not contain significant impurities of nuclear and plastid DNA.
