PREPARATION AND PURIFICATION OF E. COLI TEM-1 β-LACTAMASE

Summary

    The phage-technology for supersynthesis of β-lactamase TEM-1 in E. coli cells has been worked out. It has permited obtaining (1-2) X10⁶ units of enzyme activity per ml of cultural fluid. The method for wide-scale β-lactamase preparation is suggested, that is much simplier as against those known before. The enzyme recovery is about 45%. Our preparation is electrophoretically homogenous and may be used in immuno-enzyme assay without further purification.
